生物實驗稀釋過程分步進(jìn)行(例如 1ul母液,可以先加入1ul生理鹽水或者培養(yǎng)基,混合均勻沒有析出之后再加入2ul稀釋液,依次類推,逐漸稀釋。如果有析出及時超聲復(fù)溶。)在稀釋時要確保工作液中 DMSO 的終濃度盡量在0.1%以下,最高不要超過0.5%,并設(shè)置相應(yīng)濃度的DMSO對照組。DMSO熔點(diǎn)是18.5℃,一旦稀釋用的培養(yǎng)基溫度較低,DMSO結(jié)晶,藥物就會析出。所以稀釋過程注意一下溫度控制。
